Antioxidant Activity and Phenolic Acid Analysis of Rudbeckia laciniata var. hortensis Extract

Abstract
Polyphenols are known to be beneficial for humans as antioxidants. Especially, chlorogenic acid (CA) is one of the most available acids among all phenolic compounds. Rudbeckia laciniata var. hortensis is Rudbeckia species characterized by an abundance of CA. In this study, ethanol extract of R. laciniata var. hortensis was found to contain a large amount of polyphenols and have significant radical scavenging activity. The IC50 value for 2,2-diphenyl-1-picrylhydrazyl radical scavenging activity was 0.21 mg/mL while that for 2,2¡¯-azino-bis(3-ethylbenzothiazoline-6-sulfonate) radical scavenging activity was 0.28 mg/mL. We identified three CA isomers and three CA derivatives in R. laciniata var. hortensis using UPLC-QToF/MS. The three CA isomers were chlorogenic acid, neochlorogenic acid, and cryptochlorogenic acid, while the three CA derivatives were 3,5-dicaffeoyl quinic acid (isochlorogenic acid A), 3,4-dicaffeoyl quinic acid (isochlorogenic acid B), and 4,5-dicaffeoyl quinic acid (isochlorogenic acid C).
